Sulforaphane: What Broccoli Sprout Extract Actually Does

Sulforaphane was first isolated and characterized in 1992 by a team at Johns Hopkins led by Paul Talalay and Jed Fahey, who identified it as the most potent naturally occurring inducer of Phase II detoxification enzymes then known. It doesn't exist in intact broccoli — it forms when you chew or chop the plant, releasing an enzyme called myrosinase that converts a precursor compound, glucoraphanin, into sulforaphane. Cook the vegetable first and you destroy most of that enzyme, which is why steamed broccoli delivers a fraction of what raw or lightly cooked broccoli — and especially broccoli sprouts — can provide.

Why Sprouts Specifically

A 1997 study from the same Johns Hopkins group, published in the Proceedings of the National Academy of Sciences, found that three-day-old broccoli sprouts contained 10 to 100 times more glucoraphanin than mature broccoli heads. That finding is the entire reason broccoli sprout extract exists as a supplement category — it's a way of concentrating a compound that's inconvenient to get in meaningful amounts from food alone, since you'd need to eat several pounds of mature broccoli to match what a modest sprout serving provides.

The Mechanism: Nrf2 Activation

Sulforaphane's primary mode of action is activating the Nrf2 pathway — a transcription factor that, once triggered, switches on the production of the body's own antioxidant and detoxification enzymes, including glutathione S-transferase and NAD(P)H:quinone oxidoreductase 1 (NQO1). This is an indirect antioxidant effect: rather than neutralizing free radicals itself the way vitamin C does, sulforaphane upregulates your cells' internal defense systems, and the effect is long-lasting relative to the compound's short half-life in the bloodstream.

What the Human Trials Show

A 2012 study by Kensler and colleagues, conducted in Qidong, China — a region with high dietary exposure to aflatoxin and industrial air pollutants — found that a broccoli sprout beverage significantly increased the urinary excretion of benzene and acrolein, two toxicants tied to airborne pollution exposure, within days of daily consumption. It's one of the more direct demonstrations that sulforaphane's detoxification effects translate from cell cultures into measurable outcomes in real people.

A separate and widely cited 2014 trial published in PNAS by Singh et al. gave broccoli sprout extract to young men with autism spectrum disorder and found meaningful improvements in social interaction and aberrant behavior scores compared to placebo, with effects fading after the compound was discontinued. It's a single trial in a specific population and shouldn't be overstated, but it demonstrated a measurable neurological effect from an oral sulforaphane precursor — something that's harder to show than the detoxification data.

Bioavailability Is the Whole Game

This is where most sulforaphane products fail. Glucoraphanin alone, without active myrosinase, converts poorly — gut bacteria can do some of the conversion, but inconsistently and at low efficiency. Supplements that pair glucoraphanin with a stabilized, active myrosinase source are designed specifically to solve this, and research comparing formulations has found several-fold differences in actual plasma sulforaphane levels between products that look similar on a label. Some brands sidestep the enzyme issue entirely by delivering pre-formed, stabilized sulforaphane instead of the glucoraphanin precursor — worth checking, since the two approaches aren't interchangeable in dosing.

Dosing

Clinical trials showing measurable effects have generally used doses equivalent to 15–40mg of active sulforaphane per day, or glucoraphanin-plus-myrosinase combinations designed to yield roughly that range once converted. There's no established RDA — this isn't an essential nutrient — so "more" isn't automatically better, and most of the research has used daily, sustained dosing rather than occasional use. Thorne's Crucera-SGS is dosed at 50mg of glucosinolate equivalent per capsule and is one of the more consistently studied delivery formats in this category.

What to Look For

Check whether the label specifies active myrosinase content, not just glucoraphanin — a product listing only the precursor without confirming enzyme activity may deliver very little usable sulforaphane. Avmacol has been used in more published human sulforaphane trials than any competing brand, which is a reasonable proxy for formulation reliability. Third-party testing and clear sourcing (broccoli seed vs. sprout vs. mature plant extract) are also worth checking before buying.
