Spermidine is a naturally occurring polyamine found in every cell of the human body, with concentrations that decline steadily with age. It gets its name because it was first isolated from semen in the 1600s, but the research interest today has nothing to do with that origin — it's about a cellular cleanup process called autophagy, and a growing body of observational data linking higher spermidine levels to lower mortality.

Autophagy is the process by which cells break down and recycle damaged proteins and organelles — essentially cellular housekeeping. The foundational animal study, published in Nature Medicine, found that dietary spermidine supplementation extended lifespan in mice and reversed age-associated cardiac dysfunction through autophagy induction — an effect that disappeared when autophagy was genetically blocked, confirming the mechanism. In humans, the evidence is observational rather than interventional: a prospective population study following residents of Bruneck, Italy over 20 years found that people in the top third of dietary spermidine intake had meaningfully lower all-cause mortality than those in the bottom third, a difference the researchers estimated as roughly equivalent to 5.7 years of aging. Separate epidemiological data has linked higher spermidine intake to reduced cardiovascular and cancer mortality specifically.

It's worth being precise about what this evidence does and doesn't show. These are observational and animal findings, not large randomized controlled trials with hard endpoints in humans. Spermidine isn't the only lever for autophagy — extended fasting, caloric restriction, and exercise all activate the same general pathway, and the effects appear to be at least partly additive. Researchers studying spermidine have specifically noted it may offer a way to approximate some fasting-associated autophagy benefits without the caloric restriction itself, though this is still an early and actively studied area rather than a settled finding. For readers already doing intermittent fasting or time-restricted eating, spermidine-rich foods or a modest supplement dose is a reasonable, low-risk addition rather than a replacement for those practices.
